Now on the new layer you made take your colour (mine is the swatch above) and set the layer to screen. Start highlighting the general body of the muscle, I use the water tool (in SAI) to do this. Make sure you follow the highlighted areas you made in the shadow layer, if you don't it tends to look funny. As you can see I blurred out the hindquarters and back legs already.
